Couplisg of Acetyl Donor Systems with Cholixe Acetylase*

چکیده
Previous studies of the choline acetylation system of brain and of the head ganglion of the squid have utilized acetate, ATP,’ and CoA to generate “active acetate” (1, 2). Following the isolation of acetyl CoA by Lynen et al. (3), its identity with “active acetate” has been established by the demonstration that the acetylated coenzyme is the acetyl donor in the biological acetylation of sulfanilamide (3), choline,* and orthophosphate (4), and in the synthesis of citrate (5) and acetoacetate (6). The synthesis of acetylcholine from acetyl CoA and choline has been confirmed with a partially purified choline acetylase from the head ganglion of the squid. Experiments on the coupling of choline acetylase with various acetyl donor systems, in the presence of catalytic amounts of CoA, are also report,ed. The acetyl donors used were acetyl phosphate, pyruvate, and citrate in the presence of phosphotransacet.ylase (4), the pyruvate oxidation system (7, S), and the condensing enzyme (5), respectively. Acetyl CoA As Acetyl Donor-An experiment demonstrating the enzymatic synthesis of acetylcholine from acetyl CoA and choline is given iu Table I. The formation of acetylcholine is accompanied by the appearance of a st.oichiometric amount of sulfhydryl groups. This indicates that the choline acetylase preparation employed catalyzes the following reaction. 0 Ii (1) Co&S-&-CHa + (CH,),NyCH,-CH,OH =

The N-acetyl-D-glucosaminylphosphatidylinositol De-N-acetylase of glycosylphosphatidylinositol biosynthesis is a zinc metalloenzyme.
The de-N-acetylation of N-acetyl-D-glucosaminylphosphatidylinositol (GlcNAc-PI) is the second step of mammalian and trypanosomal glycosylphosphatidylinositol biosynthesis. Glycosylphosphatidylinositol biosynthesis is essential for Trypanosoma brucei, the causative agent of African sleeping sickness, and GlcNAc-PI de-N-acetylase has previously been validated as a drug target. Inhibition of the t...
متن کاملComparative studies of histone acetylation in nucleosomes, nuclei, and intact cells. Evidence for special factors which modify acetylase action.
We have studied the pattern of histone acetylation in intact rat hepatoma tissue culture (HTC) cells, in isolated HTC nuclei, and in chromatin prepared from these cells. The results have been compared with the histone acetylation observed in a reconstituted in vitro system consisting of a variety of purified soluble nucleosomal substrates, [3H]acetyl-CoA, and one of two different purified histo...
متن کاملMCM3AP, a novel acetyltransferase that acetylates replication protein MCM3.
The MCM proteins are essential for the initiation of DNA replication. We have isolated an MCM3-associated protein (MCM3AP) in a two-hybrid screen using MCM3. Here we demonstrate that MCM3AP is an acetyltransferase which acetylates MCM3 and that chromatin-bound MCM3 is acetylated in vivo. The MCM3 acetylase, MCM3AP, is also chromatin-bound. This study also indicates that MCM3AP contains putative...
متن کاملCholinergic enzymatic activity of cerebrospinal fluid of patients with various neurologic diseases.
The cerebrospinal fluid (CSF) of normal humans contains both true and pseudocholinesterases and choline acetyl transferase (choline acetylase) detectable by radiochemical assay. Elevations of total cholinesterase, pseudocholinesterase, and true cholinesterase occur in brain tumors, meningitis, Guillain-Barre disease, hydrocephalus, and brain abscess. However, the non-specificity and inconstancy...
متن کاملAMF-1/Gps2 binds p300 and enhances its interaction with papillomavirus E2 proteins.
The cellular protein AMF-1 (Gps2) positively modulates gene expression by the papillomavirus E2 protein (D. E. Breiding et al., Mol. Cell. Biol. 17:7208-7219, 1997). We show here that AMF-1 also binds the transcriptional coactivator p300 in vitro and in vivo. E2 interacted weakly with p300. These observations led to a model in which AMF-1 recruits p300 into a complex with E2. Cotransfection of ...
